黑果腺肋花楸胚乳愈伤组织的诱导被引量:1

[目的]研究影响胚乳愈伤组织诱导的因素,为三倍体植株再生奠定基础。[方法]以黑果腺肋花楸种子成熟胚乳为试材,研究取材时期、培养基种类、激素种类等因素对愈伤组织诱导的影响。[结果]取转色期的胚乳在MS+TDZ 0.05 mg/L+IBA 2.0 mg/L+蔗糖40 g/L+琼脂8 g/L的培养基上愈伤组织长势最好,诱导率最高可达92%,颜色为黄绿色且结构致密。[结论]适宜黑果腺肋花楸胚乳愈伤组织诱导的培养方法为转色期取材,光照强度为800 lx,培养基配方为MS+TDZ 0.05 mg/L+IBA 2.0 mg/L+蔗糖40 g/L+琼脂8g/L。

“贝吉佳”草莓脱毒苗生根及驯化研究被引量:4

[目的]研究"贝吉佳"草莓组培生根环境,并对组培苗进行驯化移栽。[方法]以"贝吉佳"草莓匍匐茎茎尖为外植体,研究其生根的影响因素及移栽基质对成活率的影响。[结果]最佳的生根培养基浓度为1/2MS,生根率可达100%;IBA的生根效果优于IAA,当IBA浓度为0.3 mg/L时生根效果最好,根粗壮,须根最多;活性炭浓度为0.5 g/L,最适合"贝吉佳"草莓根的生长。组培苗的驯化中,珍珠岩和蛭石组合基质的成活率明显高于腐殖土和沙子的基质组合,成活率最高可达98.1%(珍珠岩∶蛭石=1∶1),在各种基质上的苗均可以正常开花结果、正常抽生匍匐茎。[结论]为实现"贝吉佳"草莓脱毒苗的规模化生产和快速培育优良种苗提供技术支持。

Study on Rooting of Strawberry Bechika被引量:1

[Objective] The aim was to study rooting environment of strawberry "Bechi- ka" seedlings, and the somaclone domesticated transplanting was carded on. [Method] With the stem tip of stolon as material, the influence factom of rooting and the effects of different transplanting substrata on survival rate was investigated. [Result] The results showed that: 1/2MS was the best concentration for rooting and the rooting rate could reach 100%. IBA was better than IAA for rooting on growth regu- lator levdls, and it had the best effect on rooting with 0.3 mg/L, which produced the thickest and most-fibrous roots. Activated carbon had positive influences on the growth of roots, and the suitable concentration of activated carbon was 0.5 mg/l_ for rooting. The study found that survival rate of tissue culture seedlings was obviously higher in the combination of perlite and vermiculite than that in the combination of humus and sand, and the survival rate could reach 98.1% (perlite:vermiculite=l:l). Seedlings in various transplanting substrata could normally bloom, fruit and form stolons. [Conclusion] The study provided technical support for achieving large-scale production of virus-free strawberry and quickly cultivating good seedlings.

以黑果花楸组培苗为试材,采用石蜡切片和叶表皮撕取法,比较正常苗与玻璃化苗茎叶横切面显微结构以及叶表面气孔特征.结果表明,黑果腺肋花楸栅栏组织由2~3层细胞构成,下表皮气孔下陷,小而密集,茎内分布大量导管、筛管.玻璃化苗主脉不明显,细胞排列凌乱不规则,栅栏组织与海绵组织的区别不明显,气孔开口大近圆形,茎肿胀粗大髓腔内薄壁细胞较多、已破裂.

Comparison of Morphological Microstructure of Vitrified and Normal Shoots of Aronia melanocarpa Elloit被引量:2

The Aronia melanocarpa El oit was used as test material. The microstruc-ture of normal and vitrified shoots and the characteristics of their stomas on leaf surface were compared by paraffin section and leaf epidermis-tearing method. The results showed the palisade tissue of Aronia melanocarpa El oit consists of 2-3 lay-ers of cells. The stomas on lower epidermis cave in, and are smal and dense. There are abundant vessels and sieve tubes in stems. ln contrast, the main veins of vitrified shoots are unobvious, messy and irregular. The boundary between pal-isade tissue and spongy tissue is not obvious. The stomas open circularly and bigly. The stems are swel ing and thick, but the pith parenchyma cells are broken.
